Clongen Laboratories offers decades of molecular biology expertise for rodent genotyping.  Confirmation of the presence or absence of specific genes in transgenic and knockout animals is accomplished using PCR, RT-PCR, Southern blotting and Slot blot assays.  Existing protocols can be used or new assays can be optimized and adapted for new genotyping submissions.

Prices (Effective January 1, 2006)

There is a one time setup fee for assay transfer of new protocols and $22 for each test thereafter.  If the protocol is established and can be provided by the client, a one time $125 is required and $22 for each assay thereafter.

PCR Testing

DNA extracted from tail tissue clippings is used as the target template and the proper primers specific for the detection of either the transgene or a knockout gene in the PCR reaction mixture will result in the amplification of the expected size fragments.  Clongen Laboratories can determine the genotype of animals within your colony. The resulting amplification of primer-specified target DNA allows for the determination of the genetic nature of the tested animals with respect to gene insertion or deletion - and therefore their genotype.

  • Reporting of Results and Sample Submission

    PCR results are reported to the customer within 5-14 days following receipt of samples. Assay transfer of new assays may require an additional week.

    The most desirable samples for genotyping are tail snips.  Tail snips should be collected and immersed in 70% ethanol and refrigerated (4°C) prior to shipping. (Other tissues, such as toe clips, ear punches, muscle and brain tissue may also be used.) Samples should be shipped overnight on ice packs. Please contact Clongen Laboratories prior to shipping any samples.
     

Southern Blot

Hybridization differs from PCR in that the target DNA is not amplified.  The target sequence is detected using a signal amplification technology where a labeled probe hybridizes to the target and following stringent washing steps, the probe/target complex is detected using color development.  Southern blotting requires digestion of the genomic DNA with restriction enzymes to produce fragments of various sizes. Those fragments are run on an agarose gel and blotted onto a membrane. The membrane is hybridized with a probe designed to complement the target DNA.  Following development of the signal or image recording, the resulting banding pattern is analyzed to determine the genotype or carrier status of animals within the tested colony.

    Southern Blot results are reported to the customer within 8-20 days following receipt of samples. However, transfer and validation of new protocols may require additional time.

Slot Blot

Slot blots are the least comprehensive of the genotyping assays. While they determine the presence or absence of a transgene for transgenic mutant models (carrier versus wild-type determinations), they do not provide added confirmation of an interpretable banding pattern. Although hybridization is the method of detection for both Southern and Slot blots, DNA is applied directly to one small area on a membrane, not separated by size in Slot blotting. Transgenic model genotype information will be reported simply as carrier or wild type status.
